first : put lot of lighting to make sure the aquarium bright enough.

so u can set lower iso and higher shutter speed to capture it.

use lens 50mm f/1.4 or 50mm f/1.2 L to shoot in low light condition.

bro liubei, your lens EFS 55-250 with small aperture 4-5.6IS did not set for nite/dark situation(lowlite)..
should use bigger aperture like hendri said...50 F 1.2...1.4 or 1.8 (cheaper 700rb) this fix lens, no zooming trigger

if you still want use previous lens...set zoom at 55 so the aperture will b at 4 then set ISO to 1000 or more (depend on situation) and try not to breath while take a picture:D:D:D:D...turn on many lite would help too

iso 1600 sure not sharp. must take under iso 400 for sharper picture.

boss... is flash needed?

In my opinion, an external flash is more important. Because it offers options adjusting the position with an omnibounce to avoid a direct flash ( too harsh + reflection on glass).
Large aperture lens may help in capturing low light situation but the DOF is too shallow.
For a 40D, one could pump the ISO to 800 without losing much details with proper flash exposure.
It takes trials and errors to get it perfectly right on aquarium shooting.
Set to Manual.
Set A ranging from F6 to F8 ( smaller aperture means more details)
Set S started from 1/50s to 1/100s ( faster shutter speed means ability to freeeze motion, but u get darker background as u increase the shutter speed)
The crucial part is the position of the external flash, adjust direction until it achieve a non-reflective capture ( omnibounce required).

Not entirely accurate, while it's true that white lights ( and additional lights) help to get "clearer" picture, the limited light will still prevent u from capturing a tact sharp and detailed picture.
If one can afford a Dslr, there is no reason to save on a speedlite.
